What is the difference between the volume of the sample and the volume of the chain?

The difference between sample and chain volume comes down to your DAW or equipment. One for your sample/instrumental/plugin and one for the overall chain of that track where its placed. Your chain is essentially the volume of your track 1,2,3 and so on.
When it comes to balancing your sample you have to, first, make sure your levels are right. Make sure there is no clipping or distortion. You want to start with EQing the sample to remove impurities.

What of them do you tweak when you balance the sample?

When you are happy with how your sample sounds, then you can begin leveling for your mix always start with the sample volume first. Set it to around -12db - -18db to leave headroom. When the sample, meaning the source of the sample you can now level from the mixer section.
Here you can add effects or tweak however you like and mix only from the chain mixer faders, not from the sample volume. I hope that made sense lol.

How do you process your layered samples to sound professional?

For my Layered Samples, I mainly make sure I get it sounding as crisp as I can. This crispness comes from a good amount of highs and the right balance of mids and highs.
Once you get it sounding good, you can then add effects to make it more vintage, or you can add effects to make it appear wider or even to EQ again. Everyone has a varied definition of what sounding professional is.
